摘要
目的探讨miR-129在食管鳞癌早期诊断及肿瘤淋巴结转移中的作用。方法用Trizol提取细胞和组织中的总RNA,应用实时定量RT-PCR技术,检测miR-129在三种食管鳞癌细胞系EC9706、EC109、KYSE150及其在40对食管鳞癌组织和正常食管组织中的表达差异。结果 EC9706、EC109中miR-129的含量大于在KYSE150中的含量。miR-129在食管鳞癌组织中表达显著上调(P<0.001),平均上调倍数为(7.65±1.06)。miR-129表达上调水平与食管鳞癌临床分期(P=0.005)及淋巴结转移(P=0.002)存在相关性。结论在不同食管鳞癌细胞系和组织中的表达差异提示:miR-129在不同人种间存在差异表达及其可能参与了食管鳞癌的发生发展过程,可作为一个潜在的食管癌生物标记。
Objective To explore the microR-129 expression patterns associated with the early diagnosis and the lymph node metastasis of esophageal squamous cell carcinoma(ESCC) .Methods Total RNA isolation was done with Trizol.Quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR) was performed for miR-129 expression in cells and tissues.Results The content of miR-129 was identified as higher expressed in EC9706 and EC109 of China than in KYSE150 of Japan.The expression level of miR-129 was higher in the tumor tissues than that in the normal esophageal tissues(P0.001) with average up-regulated level of(7.65±1.06) .And high level expression of miR-129 was significantly correlated with advanced clinical stage(P=0.005) ,lymph node metastasis(P=0.002) .Conclusions The altered expression of miR-129 might play a role in the pathogenesis and progression of ESCC,and it may be a useful molecular marker for esophageal carcinoma.Furthermore,the expression levels of miR-129 maybe exist a gap in different races.
