摘要
目的建立中药复方制剂感冒软胶囊(羌活、麻黄、桂枝、葛根、黄芩、川芎、防风等)中盐酸麻黄碱、盐酸伪麻黄碱、葛根素及黄芩苷4种活性成分同时测定的胶束电动毛细管色谱法。方法以甲醇提取样品,电泳缓冲液为10 mmol/L磷酸氢二钠-20 mmol/L硼酸(pH 8.0),内含20 mmol/L十二烷基硫酸钠(SDS)及体积分数分别为27%(v/v)异丙醇和18%(v/v)乙腈作有机改性剂,采用75μm(i.d)×60(ef 50)cm的未涂层弹性石英毛细管柱0,.5 psi(进样时间6 s)压力进样,分离电压30 kV,紫外检测波长214 nm,温度25℃。结果 4种活性成分在23 min内获得基线分离,盐酸麻黄碱、盐酸伪麻黄碱、葛根素和黄芩苷的线性范围分别为1.5~200μg/mL(r=0.999 4)、2.4~150μg/mL(r=0.999 8)、1.8~120μg/mL(r=0.999 1),4.5~300μg/mL(r=0.999 1),检出限分别为0.50、0.800、.60、1.5μg/mL(S/N=3)。样品加标回收率在91.0%~109%之间,相对标准偏差均小于3.2%。结论本法简便、快速,具良好的精密度和回收率,已成功用于实际样品的分析。
AIM To establish a new method for simultaneously determing the contents of ephedrine hydrochloride,pseudoephedrine hydrochloride,puerarin and baicalin in Ganmao Soft Capsule(Notopterygii Rhizoma et Radix,Ephedrae Herba,Cinnamomi Ramulus,Puerariae Lobatae Radix,Scutellariae Radix,Chuanxiong Rhizoma,Saposhnikoviae Radix,etc.) by micellar electrokinetic capillary chromatography(MECC).METHODS The samples in preparation were extracted by methanol,20 mmol/L SDS-10 mmol/L Na2HPO4-20 mmol/L H3BO3mixture(containing 27% 2-Propanol and 18% acetonitrile,pH=8.0) was selected for the running buffer.The four effective components were separated by an uncoated fused silica capillary(75 μm×60 cm) with the detection wavelength of 214 nm,voltage of 30 kV and temperature of 25 ℃.RESULTS The baseline separation of four components was achieved within 23 min.The linear ranges were 1.5-200 μg/mL,2.4-150 μg/mL,1.8-120 μg/mL,4.5-300 μg/mL for ephedrine hydrochloride,pseudoephedrine hydrochloride,puerarin and baicalin with detection limits of 0.50,0.80,0.60,1.5 μg/mL,respectively.The average recoveries of the four components were between 91.0%-109% with all relative standard deviations less than 3.2%.CONCLUSION The method is simple,rapid with excellent precision and high recovery and can be used for the quality control of Ganmao Soft Capsules.
