摘要
目的考察戊二醛(GA)交联的黏质沙雷氏菌ECU1010脂肪酶活性包涵体的酶学性质。方法分别对游离酶和固定化酶的pH稳定性、热稳定性、Km值、有机溶剂和金属离子耐受性进行研究,同时考察固定化酶的操作稳定性和贮存稳定性。结果固定化酶在pH 5.0~10.0稳定性较好;60℃以下仍可保持48%以上的酶活;对亲水性有机溶剂和金属离子的耐受性增强;操作稳定性和贮存稳定性大有提高。结论脂肪酶活性包涵体经固定化后其稳定性显著提高。
OBJECTIVE To investigate the enzymatic properties of glutaraldehyde-immobilized lipase active inclusion bodies from Serratia marcescens ECU1010. METHODS Comparative studies on the enzymatic properties of free and immobilized enzymes were conducted. The main properties studied in this work included the stability of temperature and pH, as well as in the presence of va- rious organic solvents and metal ions. Moreover,the operation stability and storage stability were further investigated. RESULTS The immobilized lipase was stable over a broad pH range of 5.0 - 10. 0 and remained over 48% of its activity when the temperature was below 60℃. The tolerance to water-miscible organic solvents and metal ions was raised. The operation stability and storage stability were highly improved. CONCLUSION The stability of immobilized lipase active inclusion bodies is significantly improved.
