摘要
目的研究香青兰有效部位滴丸对犬急性心肌缺血损伤的保护作用。方法犬冠状动脉结扎致心肌缺血模型,香青兰有效部位滴丸低(25 mg.kg-1)、中(50 mg.kg-1)、高(100 mg.kg-1)剂量组十二指肠给药后采用心外膜电图标测心肌损伤程度(ΣST)及损伤范围(NST),采用红四氮唑(TTC)染色法测定心肌梗死区域。测定血清中天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)活性。检测香青兰有效部位滴丸对实验动物心肌耗氧量、血压、心率(HR)及冠脉流量(CF)及冠脉阻力(CR)的影响。结果犬急性心肌缺血形成后,十二指肠给予香青兰有效部位滴丸50、100mg.kg-1可改善心肌缺血程度,并缩小心肌缺血范围,减少梗死心肌的范围;高剂量还降低心肌耗氧量,抑制CK-MB活性的升高。100 mg.kg-1可降低血压,但对HR、CF、CR无明显影响。结论香青兰有效部位滴丸对犬冠脉结扎所致心肌缺血性损伤具有一定保护作用。
OBJECTIVE To study the effect of the dripping pills of moldavica effective composition on acute ischemic myocardial injury induced by ligation of coronary artery in dogs. METHODS Myocardial ischemia was induced by ligation of the left coronary anterior descending artery in anaesthetized thoraco-opened dogs. The dripping pills of moldaviea effective composition at 25, 50 and 100 mg. kg-1 were administrated into dodeeadactylon. Epieardial electrocardiogram and quantitative histological analysis combined with red tetrazoline (TYC) staining were used to determine the range and degree of the damage induced by myocardial ischemia. The activities of serum AST, LDH, and CK-MB were measured. The effects of dripping pills on blood pressure, heart rate (HR) , coronary artery flow (CF) and coronary artery resistance (CR) were observed. RESULTS 50, 100 mg · kg-1 dripping pills significantly decreased the severity of myocardial ischemia and the area of myocardial infarction. The dripping pills at high dose obviously reduced serum CK-MB activity and myocardial consumption of oxygen. In addition, effective composition of moldavica dripping pills lowed the blood pressure without significant influence on HR, CF, and CR. CONCLUSION The dripping pills of moldavica effective composition can protect anesthetized thoraco-opened dogs from ischemic myocardial injury induced by ligation of coronary artery.
