摘要
【目的】建立冬种马铃薯试管苗高效生产技术体系。【方法】在不同植物生长调节剂、光强和昼夜温差、培养器皿和封口材料、固液体MS和1.5MS培养基条件下对马铃薯试管苗进行壮苗培养。【结果】在MS培养基中添加B9和CCC,均对马铃薯试管苗壮苗培养有促进作用,但以B9的效果最佳、最持久;当光照强度为3000lx、昼夜温差为8℃左右时,即培养温度在18~26℃时壮苗培养效果较佳。与固体培养基相比,1.5MS液体培养基对试管苗的壮苗效果更明显;与试管和250mL罐头瓶相比,透光、透气性好的150mL三角瓶带塑料封口膜更有利于马铃薯试管苗壮苗培养,且操作方便,试管苗生长较好。【结论】在光照强度3000lx、温差为8℃条件下,在带塑料封口膜的150mL三角瓶中采用添加B9的1.5液体MS培养基对马铃薯试管苗进行壮苗培养,可获得较好的培养效果。
[Objective]The experiment was aimed to establish highly efficient production technology system for test tube potato plantlets. [Method ] The effects of different cultural conditions, viz., plant growth regulators, light intensity, temperature difference between day and night, culture container and medium were observed on the growth and vigour of potato plantlets. [Result]The test tube potato plantlets showed better growth by addition of damiuozide (B9) in the media compared to addition of eycoeel (CCC). The test tube plantlets grew strongly under conditions of 3000 lx light intensity and about 8℃ temperature difference between day and night (18-26℃). Compared to the solid culture medium, 1.5MS liquid medium showed better culture effects on test tube plantlets. It was observed that test tube plantlets grew well when cultured in 150 mL transparent flasks with plastic sealed film compared to 250 mL cans. [Conclusion lit is concluded that best test tube potato plantlets may be cultured in flask sealed with plastic film using 1.SMS liquid culture medium supplemented with B9 under conditions of 3000 lx light intensity and about 8℃ temperature difference between day and night (18-26℃).
