摘要 目的探讨Omi/HtrA2和caspase-7在喉鳞状细胞癌中的表达及相关性。方法采用免疫组织化学方法检测40例喉鳞癌组织(喉癌组)和10例正常喉黏膜组织(对照组)中Omi/HtrA2和caspase-7表达,采用图像分析系统对表达结果进行定量分析,并分析其与临床病理特征相关性。结果喉鳞状细胞癌组织Omi/HtrA2表达阳性率显著高于正常喉黏膜组,两组比较差异有统计学意义(P<0.05),其阳性单位分别为26.952±7.145和10.395±6.978,差异有统计学意义(P<0.05);喉鳞状细胞癌组织caspase-7表达的阳性率高于正常喉黏膜组织,其阳性单位分别为29.365±8.152和9.362±4.234,差异有统计学意义(P<0.05)。喉鳞状细胞癌组织Omi/HtrA2和caspase-7表达的阳性单位与年龄、性别、临床分型(肿瘤部位)、临床TNM分期和淋巴结转移无关,与病理分级(G1→G2+G3)有关(P<0.05)。喉鳞状细胞癌组织中Omi/HtrA2与caspase-7表达有相关性(r=0.332,P<0.05)。结论 Omi/HtrA2和caspase-7可通过调节细胞凋亡,对喉癌的发生和发展起重要作用。 Objective To investigate the expression of Omi/HtrA2 and caspase-7 and their relationship in laryngeal squamous cell carcinoma(LSCC). Methods The expression of Omi/HtrA2 and caspase-7 protein in 40 cases of LSCC tissues and 10 cases of normal laryngeal mucous membrane tissues was detected using immunohistochemical method and quantitative analysis of image.The association of Omi/HtrA2 and caspase-7 expression with clinical pathological features was analyzed. Results The positive rates of Omi/HtrA2 and caspase-7 expression were significantly higher in LSCC tissues than that in normal laryngeal mucous membrane tissues,respectively.The positive unit(PU) of expression of Omi/HtrA2 protein was 26.952±7.145 in LSCC tissues,and 10.395±6.978 in normal laryngeal mucous membrane tissues(P0.05).The positive unit(PU) of expression of caspase-7 protein was significantly higher in LSCC tissues than in laryngeal mucous membrane tissues(29.365±8.152 vs 9.362±4.234,P0.05).In LSCC tissues,no correlation was found between positive unit(PU) of Omi/HtrA2 and caspase-7 expression and sex,age,tumor site,TNM stage and lymphatic matastasis,but there was significant correlation between positive unit(PU) of Omi/HtrA2,caspase-7 expression and differentiation grading(G1→G2+G3)(P0.05).The expressions of Omi/HtrA2 was correlated with caspase-7 in LSCC tissues(r=0.332,P0.05). Conclusion Omi/HtrA2 and caspase-7 may play important roles in the occurrence and development of LSCC by regulating the apoptosis of cancer cells.
