摘要
为开发蚕蛹新资源,测定了桑蚕蛹中13种氨基酸含量。采用OPA柱前衍生化,反相高效液相色谱法,梯度洗脱方法,荧光检测器检测,分析蚕蛹中各种氨基酸含量。蚕蛹中天冬氨酸、谷氨酸、丝氨酸、组氨酸、甘氨酸、苏氨酸、丙氨酸、牛磺酸、蛋氨酸、缬氨酸、苯丙氨酸、异亮氨酸、亮氨酸的含量分别为63.97mg/g、72.48mg/g、32.46mg/g、52.21mg/g、6.531mg/g、35.63mg/g、31.37mg/g、21.39mg/g、1.806mg/g、71.01mg/g、68.08mg/g、35.86mg/g、25.34mg/g,总氨基酸含量为518.15mg/g,其中人体必需氨基酸占45.88%。建立的反相高效液相色谱法测定微量氨基酸的方法简便、快速、灵敏、准确。
The aim was to develop new resources about silkworm chrysalis, 13 amino acids were determined in silkworm pupa. The contents of amino acids were analyzed by column derivatization with OPA, reversed phase high performance liquid chromatography (RP-HPLC), gradient elution method and fluorescence detection. The contents of aspartic acid, glutamic acid, serine, histidine, glycine, threonine, alanine, taurine, methionine, valine, phenylalanine, isoleucine and leucine contents were 63.97 mg/g, 72.48 mg/g, 32.46 mg/g, 52.21 mg/g, 6.531 mg/g, 35.63 mg/g, 31.37 mg/g, 21.39 mg/g, 1.806 mg/g, 71.01 mg/g, 68.08 mg/g, 35.86 mg/g and 25.34 mg/g, respectively. Total amino acid content was 518.15 mg/g with 45.88% essential amino acids. RP-HPLC method for determining amino acids is simple, rapid, sensitive and accurate.
