微孔板发光二极管诱导荧光-表面活性剂增敏法测定盐酸小檗碱 被引量:3

Determination of Berberine Hydrochloride by Light-emitting Diode Induced Fluorescence Microplate Analyzer with Surfactant Sensitization

摘要 研制了微孔板发光二极管诱导荧光分析仪,以发光二极管为激发光源、Y形光纤束传导激发光和荧光,采用平面型光敏二极管接收荧光。采用二维定位器移动微孔板,可实现对多个样品快速连续的测定。利用弱碱性介质中阴离子表面活性剂十二烷基苯磺酸钠(SDBS)对盐酸小檗碱荧光的增敏作用,在pH 9.0的Britton-Robinson缓冲溶液中,以SDBS为增敏剂,以470nm的发光二极管为光源,测定盐酸小檗碱的含量。结果显示,盐酸小檗碱浓度在5.0~120mg/L范围内的相关系数为0.9987,检出限为1.3mg/L,RSD为2.4%。将此仪器应用于制剂与中药材中盐酸小檗碱含量的测定,加标回收率为96.5%~103.5%。 A light-emitting diode induced fluorescence microplate analyzer was presented by using a light-emitted diode as excitation source,a planar photodiode as detector.The excitation light and fluorescence signal were transmitted by a Y-shape optical fiber bundle.All operation procedures of sample exchange could carry out by the movement of a 2-D localizer in a short time.In a Britton-Robinson buffer solution(pH 9.0),the fluorescence of berberine could be enhanced by the presence of anion surfactant.Under the excitation of LED of 470 nm,the linear range,limit of detection and relative standard deviation of berberine in pharmaceutical preparation were 5.0-120 mg/L,1.3 μg/L and 2.4%,respectively.This equipment was applied to determine berberine hydrochloride in pharmaceutical preparation and Chinese medicinal herb with the recoveries of 96.5%-103.5%.
