摘要
目的观察远志水提物对D-半乳糖和亚硝酸钠所致阿尔茨海默病模型小鼠学习记忆功能的影响,初步探讨其可能的机制。方法将60只小鼠随机分成6组,即空白对照组、模型组、脑复康组和远志水提物高、中、低剂量组,每组10只。腹腔注射D-半乳糖和亚硝酸钠建立阿尔茨海默病模型,同时灌胃给予不同剂量的远志水提物4周,利用Morris水迷宫评价学习记忆功能后测定血清超氧化物岐化酶(SOD)活性和丙二醛(MDA)含量。结果与模型组比较,远志水提物高剂量组小鼠搜索站台的潜伏期均明显缩短(P<0.05),高、中剂量组在原站台所在象限停留时间明显延长(P<0.05),远志水提物高剂量组小鼠血清中SOD活性明显增加(P<0.01),高、中剂量组小鼠血清中MDA含量明显降低(P<0.01)。结论远志水提物能预防和改善D-半乳糖和亚硝酸钠所致阿尔茨海默病模型小鼠学习记忆功能减退,其机制可能与抗氧化作用有关。
Objective To observe the effects of polygala tenuifolia extracts(PTE) on learning and memory abilities in Alzeimer's disease mice,and its possible mechanism.Methods 40 male rats were randomly divided into 6 groups:normal control,model control,Naofukang treatment,and three doses of PTE groups,each with ten rats.Alzeirmer's disease model was induced by i.p.injection of D-galactose and sodium nitrite,and PTE groups were treated with i.g.PTE for 4 weeks.The learning and memory abilities were detected by monitoring its behavior in Morris water maze,and measured the activities of serum T-SOD and the contents of MDA in serum.Results Compared with model control group,high dose of PTE significantly shortened the platform searching latency(P〈0.01),middle and high dose of PTE prolonged the retention time at the original platform(P〈0.05),high dose of PTE increased the serum T-SOD(P〈0.01),and middle and high dose of PTE decreased the content of MDA significantly(P〈0.01).Conclusion PET improves the abilities of learning and memory of D-galactose and sodium nitrite induced Alzeimer's disease mice,which might contribute to its antioxidation function.
