摘要
以菌丝生物量、胞内多糖含量及胞内多糖产量为评价指标,对鲍氏针层孔菌液体深层发酵培养的培养基配方及发酵条件进行了优化。通过单因素实验和正交实验筛选液体发酵培养基,结果表明,最适培养基为:玉米粉4%,麸皮2%,KH2PO40.4%,MgSO4·7H2O0.05%。进一步对培养条件进行优化,得到最适生长的液体发酵条件为:培养温度28℃、pH6.5、接种量12%、装液量150mL/500mL、转速140r/min、发酵时间168h。在此优化工艺条件下,菌丝生物量、胞内多糖含量及产量由优化前的12.837g/L,15.930mg/g,204.493mg/L分别提高到19.410g/L,20.935mg/g,406.348mg/L。
Taking the mycelial biomass,the content of intracellular polysaccharide,and the yield of intracellular polysaccharide as detecting index,the optimal submerged fermentation parameters of P.baumii were investigated by single factor test and orthogonal test.The results showed that the optimal culture was corn flour 4%,bran 2%,KH2PO4 0.4%,and MgSO4·7H2O 0.05%.The best fermented condition was culture temperature 28℃,pH6.5,inoculation volume 12%,the amount of liquid 150mL/500mL,rotation speed 140r/min,and fermentation time168h.Before technology was optimized,mycelial biomass and content and yield of intracellular polysaccharide was 12.837g/L,15.930mg/g,204.493mg/L.But mycelial biomass and content and yield of intracellular polysaccharide attain 19.410g/L,20.935mg/g,406.348mg/L respectively under the optimum condition.
