摘要
从亨氏马尾藻中提取岩藻聚糖硫酸酯(SPF),采用超声波辅助水提法,以多糖提取率为指标,利用Statistica软件分析多糖提取率与各因素的相关性,再通过响应面法的中心组合设计优化SPF的工艺参数。建立了多糖提取率与超声时间、提取温度和提取时间之间的二次回归模型,即:Y=10.40+0.52A-0.047B+0.32C-0.28AB+0.13AC-0.12BC-0.52A2-0.13B2-0.46C2。优化后的最佳提取条件为:提取pH6.00,提取温度80℃,提取料液比1:30,提取时间3.57h,超声时间48.5min,超声功率350W。SPF得率可达10.32%,SPF中总多糖含量为36.65%,岩藻糖含量为7.35%,硫酸根含量为13.60%。
Fucoidan from Sargassum hemiphyllum was extracted by ultrasonic wave extraction method.Statistica and response surface methodology(RSM)were used to optimize the SPF parameters.The results showed that the behavior of the system was explained by the following quadratic equation between SPF and the factors(ultrasonic time,extraction temperature and extraction time):y=10.40+0.52A-0.047B+0.32C-0.28AB+0.13AC-0.12BC-0.52A2-0.13B2-0.46C2.SPF rate condition was extracted based on that equation:extraction time 3.57h,80℃ and ultrasonic time 48.5min.The last optimization condition:pH6.00,temperature 80℃,the ratio of solution to algae power 1:30,extraction time 3.57h,ultrasonic time 48.5min,the power 350W,and the SPF extraction rate was 10.32%,polysaccharide content 36.65%,fucose content and sulfate group content were 7.35% and 13.60%.
