表达小反刍兽疫病毒H基因重组腺病毒的构建及对小鼠免疫原性的试验 被引量:2

摘要 人工合成密码子优化的小反刍兽疫病毒(PPRV)H基因,将其克隆到腺病毒穿梭载体pShuttle-CMV中,电转化BJ5183感受态细胞,获得重组腺病毒质粒pAdV-H,转染AD-293细胞,获得重组腺病毒rHAdV-H。用PCR和Dot-ELISA鉴定H基因在AD-293细胞中的表达,同时用一步生长曲线,测定该重组腺病毒的复制能力。接种小鼠评价免疫效力,用间接ELISA试验检测抗体水平,结果表明,PPRV H基因在AD-293细胞中获得了表达,子代重组腺病毒在感染细胞后60 h滴度最高,为3×108PFU/mL。rHAdV-H免疫小鼠产生针对PPRV特异性抗体,表明该重组腺病毒可以诱导抗PPRV的特异性免疫应答。
