卵巢癌中OPN、αvβ3和VEGF的表达与侵袭转移的关系 被引量:2

Expression of OPN, αvβ3 and VEGF in EOC and their relationship with invasion and metastasis

摘要 目的探讨骨桥蛋白(OPN)、整合素αvB3和血管内皮生长因子(VEOF)在上皮性卵巢癌(EOC)中的表达及其临床意义,寻求卵巢癌可能的肿瘤标记物。方法采用免疫组化方法,检测50例卵巢癌组织、20例卵巢良性肿瘤组织及10例正常卵巢组织标本中骨桥蛋白、整合素αvβ3和血管内皮生长因子的表达,分析它们与患者临床病理参数的关系及其之间的相互关系。结果骨桥蛋白、整合素αvβ3和血管内皮生长因子在卵巢癌组的表达率均明显高于正常组和良性肿瘤组(X^2值分别为41.302、28.544、28.848,均P〈0.05)。在晚期卵巢癌中的骨桥蛋白、整合素αvβ3和血管内皮生长因子阳性表达率均高于早期的卵巢癌(X^2值分别为7.728、23.000、9.432,均P〈0.05);分化差卵巢癌中骨桥蛋白和整合素αvβ3阳性表达率明显高于分化好的卵巢癌(X^2值分别为6.542、12.167,均P〈0.05);有淋巴结转移的卵巢癌中,骨桥蛋白、整合素αvβ3和血管内皮生长因子阳性表达率高于无淋巴结转移的卵巢癌(X^2值分别为6.629、13.782、4.726,均P〈0.05)。骨桥蛋白与整合素αvβ3、血管内皮生长因子在卵巢癌中的表达均显著相关(,值分别为4.973、5.821,均P〈0.05)。骨桥蛋白、整合素αvβ3和血管内皮生长因子的共表达与卵巢癌的淋巴结转移显著相关(X^2=9.343,P〈0.05)。结论骨桥蛋白、整合素αvβ3和血管内皮生长因子的过度表达可能促进了卵巢癌的浸润转移,骨桥蛋白表达与整合素αvβ3、血管内皮生长因子表达分别具有正协同作用。联合检测骨桥蛋白、整合检测骨桥蛋白、整合素αvβ和血管内皮生长因子可作为预测卵巢癌浸润转移及评价患者预后的生物学指标。 Objective To investigate the expression of osteopontin ( OPN), integrin αvβ3 and vascuoar endothelial growth factor (VEGF) in epithelial ovarian carcinoma (EOC) and their clinical significance, so as to seek possible tumor markers for EOC. Methods Immunohistochemistry stain was used to detect the expression of OPN, αvβ3 and VEGF in 50 patients with ovarian cancer, 20 patients with ovarian benign tumor and 10 cases of normal ovarian tissue, and their interrelationship and the association with clinical pathologic factors were analyzed. Results The expression of OPN, αvβ3 and VEGF in patients with EOC was significantly higher than that in patients with ovarian benign tumor and normal ovarian tissue (X^2 was 41. 302, 28. 544 and 28. 848 respectively, all P 〈 0.05). The positive expression rate of OPN, αvβ3 and VEGF was significantly higher in patients with advanced stages of EOC than that in the patients with early stages(X^2 was 7. 728 ,23. 000 and 9. 432 respectively, all P 〈 0.05 ). The positive expression rate of OPN and αvβ3 protein was significantly higher in EOC of poor differentiation than that of good differentiation (X^2 was 6. 542 and 12.617 respectively, both P 〈 0.05 ). Compared with the positive expression rate of OPN, αvβ3 and VEGF protein in EOC without lymde metasis, it was higher in EOC with lymde metasis (X^2 was 6.629,13. 782 and 4. 726 respectively, all P 〈 0.05 ). The expression of OPN showed direct correlation with αvβ3 and VEGF in EOC (X^2 was 4. 973, 5. 821, P 〈0.05). The co-expression of OPN, αvβ3 and VEGF had remarkable correlation to lymde matasis of EOC (X^2 = 9. 343, P 〈 0.05 ). Condusion The over-expression of OPN, αvβ3 and VEGF may promote the invasion and metastasis of ovarian carcinomas. The expression of OPN has positive cooperativity with the expression of αvβ3 and VEGF. Combined detection of OPN, αvβ3 and VEGF may be used as biological indicator to predict the invasion and metastasis of ovarian cancer and to estimate prognosis.
