摘要
以赤霉酸回收率为指标,对固相萃取的淋洗剂与洗脱剂进行了探讨;赤霉酸高效液相色谱检测波长为210nm,以保留时间、分离度、拖尾因子、谱图峰型、相对标准偏差等为指标,分别考察了流动相甲醇与水的体积比、流动相pH、流动相流速及柱温对大米中赤霉酸检测结果的影响。结果显示,固相萃取不使用淋洗剂,洗脱剂为1mL甲醇时,回收率96.67%;HPLC条件为C18柱,柱温40℃,流动相为体积比为30/70的甲醇-水溶液(pH4.0),流速为1.0mL/min。
Studied the eluting reagent in SPE,by the index of the recovery of GA3,and on the basis of the wavelength 210 nm,studied the ratio of methanol and water,the pH value of mobile phase,flow rate and column temperature on the determination of GA3 in rice by HPLC,by the index of retention time,resolution,tailing factor,peak,and RSD et al.The result showed:in the solid phase extraction,the eluting reagent was methanol(1 mL);the optimum chromatographic conditions of HPLC were:C18 column,column temperature 40 ℃,mobile phase:CH3OH/H2O=30/70(pH 4.0),flow rate 1.0 mL/min.
