摘要
【目的】分析新疆糜子种质资源遗传多样性和亲缘关系,为选育糜子优质种质资源提供理论依据。【方法】采用ISSR分子标记,对来自新疆3个地区的25份糜子样品的遗传多样性进行分析,并与来自陕西的5份样品进行比较,探讨其亲缘关系。【结果】用筛选出的13条多态性高、重复性好的引物,分别对供试材料的基因组进行扩增,共获得147条清晰可辨的谱带,其中多态性带134条;多态性比率为91.16%,Nei’s基因多样性指数为0.195,平均Shannon信息指数为0.323,遗传分化系数Gst=0.255 7;采用UPGMA法对4个不同来源的糜子材料的聚类分析表明,新疆的糜子聚为一支。【结论】研究材料的遗传变异主要存在于群体内,来自新疆的糜子材料与来自陕西的存在一定的遗传分化。研究从分子水平上为加强对糜子种质资源的挖掘、优良品种的选育及杂交亲本的选配等提供参考依据。
【Objective】 The purpose of this program is to provide theoretical basis for the breeding of high quality by investigating the genetic diversity of Panicum miliaceum in Xinjiang and their relationship.【Method】 The genetic diversity and relationship of 25 materials of Panicum miliaceum from three areas in Xinjiang and five from Shanxi were analyzed by using inter-simple sequence repeat(ISSR) markers.【Result】147 bands were amplified by 13 informative and reliable primers,of which 134 were polymorphic loci.The percentage of polymorphic loci(PPL) was 91.6%,Nei's genetic diversity(h)was 0.195,Shannon's Information index(I)was 0.32 3.UPGMA method was used to analyze the Millet cluster from four different sources.And the finding is that they all belong to one group.【Conclusion】 These results show that the main genetic variation existed within the groups and there is some genetic differentiation between the ones in Xinjiang and the one from Shanxi.They can be applied to strengthen the mining of millet germplasm at the molecular level,and provide some materials for their breeding and matching of hybrid parents in the future.
