摘要
目的:考察不同提取方法的当归补血汤对化疗贫血模型小鼠外周血细胞计数、骨髓IL-2浓度、骨髓和肾组织EPO浓度、肾组织TGF-β1浓度的影响,选取具有最佳生物效应的提取工艺。方法:采用环磷酰胺建立化疗贫血模型,正常组、模型组胃饲蒸馏水,各给药组以不同萃取方法制备的当归补血汤胃饲小鼠,连续14天。血细胞计数仪测定外周血细胞计数,双抗体夹心ELISA法检测骨髓白介素2(IL-2),肾组织EPO,骨髓EPO和骨髓TGF-β1浓度。结果:不同萃取方法制备的当归补血汤均提高小鼠外周血细胞计数(P<0.05),提高骨髓IL-2浓度,肾组织EPO浓度和骨髓EPO浓度(P<0.05),降低骨髓TGF-β1浓度(P<0.05),其中氯仿组优于其他给药组。结论:不同萃取方法制备的当归补血汤对环磷酰胺所致骨髓抑制有不同程度的保护作用,以氯仿组效果最优。推测当归补血汤中有效物质在氯仿萃取液中含量最多,生物效应最佳,为筛选该方的提取工艺提供实验依据。
Objective: To investigate the effect of Danggui Buxue Decoction extracted by different methods on cell counting of peripheral blood,IL-2,erythropoietin(EPO)and TGF-β1 concentration of the bone marrow and the EPO of the kidney,in anemia model mice induced by chemotherapy,in order to select the best biological effects of extraction process.Methods: The mice anemia model were established by cyclophosphamide chemotherapy.The mice in normal group,model group were by gavage given distilled water,and those in each treatment groups were given Danggui Buxue Decoction extracted by different methods for 14 days.Peripheral blood counting instrument was used to count blood cell,the ELISA was used to test concentrations of IL-2,EPO and TGF-β1 of the bone marrow and the EPO concentration of the kidney.Results: Danggui Buxue Decoction extracted by different methods all increased cell counting of peripheral blood(P〈0.05)and IL-2,EPO and TGF-β1 concentration of the blood marrow and the EPO concentration of the kidney(P〈0.05),but reduced TGF-β1(P〈0.05),among them the effect of chloroform group was better than those of other drug groups.Conclusion: Danggui Buxue Decoction extracted by different methods can all have protection effect in different degrees to anemia model mice induced by chemotherapy,however the effect of the decoction extracted by chloroform group is better.The possible mechanism of Danggui Buxue Decoction may be that it increases the marrow IL-2 concentration so as to protect the medullary hematopoiesic function.It is speculated that the effective substance Danggui Buxue Tang chloroform extract in the most abundant,the best biological effects,in order to select the extraction process of experimental evidence.
