摘要
目的研究海洋链霉菌SIIA-A02191产生的抗菌成分。方法用有机溶剂萃取发酵液,采用减压快速层析和制备HPLC分离纯化活性化合物,根据化合物的理化性质、紫外光谱、红外光谱、质谱、一维和二维核磁共振谱的测定数据,解析并确证化合物的结构。结果化合物1为xanfllolipin的溴代类似物,命名为bromoxantholipin,具有抗MRSA等革兰阳性细菌的强活性,MIC值范围为0.06~0.25μg/mL。结论化合物1为一新的多环口山酮类抗生素。
Objective To investigate the antibacterial constituents from cultures of the actinomycete Streptomycesflavogriseus SIIA-A02191. Methods Active compound was separated from the fermentation broth by solvent extraction and purified by vacuum flash chromatography and preparative HPLC. Its structure was elucidated on the basis of the physico-chemical properties extensive spectroscopic analysis including UV, IR, MS, 1D and 2D NMR experiments. Results Compound 1 named bromoxantholipin was elucidated to be a bromo analog of xantholipin. It showed potent antibiotic activity against Gram-positive bacteria including MRSA with the MIC of O.06-0.25μg/mL. Conclusion Compound 1 is a new polycyclic xanthone antibiotic.
