摘要
通过筛选的 1 0个随机引物 ,利用 RAPD技术 ,对丹顶鹤及灰鹤的池 DN A进行了多态性研究。结果表明 :1 0个引物共产生条带 1 3 4个 ,扩增产物片段长度一般为 3 1 0 bp~ 2 .1 kb;利用的引物不同 ,鹤种间的特异性条带也不同 ,从而可以分析鹤种间的遗传差异。根据池 DN A多态性分析 ,两鹤种间的相似性指数为 0 .2 2 3。说明
Genetic constitution of population in red crowned cranes and common cranes were analysed by RAPD technique, and 10 selected random primers were used to study DNA polymorphism of pooled DNA in the 2 species. The results indicated that 134 bands were brought about by 10 random primers, and fragment lengths of amplified products were generally from 310 bp to 2.1 kb. Every species has specific bands respectively because of different primers, which may research genetic difference among species. The similar index between Red-crowned and Common cranes according to pooled DNA was 0.223. In conclusion, it was feasible and efficient that RAPD technique was used as assistant means in analysing genetic constitution of population.
