清热解毒液对高脂血症大鼠内皮素影响的研究 被引量:30

摘要 目的和方法:应用内皮细胞铺片和免疫组化技术相结合的方法,在高脂血症大鼠主动脉内皮细胞铺片上,定量分析内皮素(ET) 的合成和释放与动脉内皮损伤的关系,观察具有抗蛇毒作用的清热解毒液对血管内皮是否有保护作用。结果:高脂血症大鼠血浆ET 水平均明显升高,内皮细胞释放内皮素增多。内皮细胞铺片显示:内皮细胞排列紊乱,细胞数量增多,胞核大小不一、染色深浅不一;IgG、ET 阳性率均高于正常对照组。可见高脂血症造成内皮细胞损伤,并使内皮细胞合成内皮素增多。中药治疗组大鼠血浆ET 水平、内皮细胞铺片IgG 和ET 阳性率均低于高脂对照组,而血脂并没有降低。结论:清热解毒液在血脂不降低的情况下,可能通过减少内皮素的合成与释放,起到保护内皮细胞的作用。 AIM and METHOD:Using immunohistochemical technique in “en face” preparations of arterial endothelium, this project is to analyse quantitatively the relationship between synthesis and release of endothelin (ET) and the endothelium injury caused by hyperlipidemia, and to observe whether heat-clearing and detoxifying fluid can protect endothelium from injury in hyperlipidemia rats with its effect on synthesis and release of endothelin. RESULTS: (1) The plasma endothelin in hyperlipidemia group (HG) increased significantly versus normal group (NG). The positive rate of IgG-and ET-positive cells were all significantly higher than those of NG. It is suggested that the model of hyperlipidemia was successful in rats, and the hyperlipidemia could damage the endothelium and increase the synthesis and release of endothelin. (2) The plasma endothelin and the positive rate of IgG-and ET-positive cells in Chinese medicine treated group (CMTG) were all decreased significantly versus HG. CONCLUSION:The heat-clearing and detoxifying fluid could protect endothelium by directly decreasing the synthesis and release of endothelin without reducing serum lipid.
