摘要
研究丙型肝炎病毒NS5A区插入序列(IS) 因子及其来源。以RT-PCR法扩增HCV NS5A区第6871至7121 区段, 对扩增产物进行核苷酸测序并与HCV-J株相应区段序列相比较。结果发现HCV NS5A 区插入一个48 bp 的IS因子, 将IS因子与HCV-J株全核苷酸序列进行同源性对照, 发现该片段来源于HCV E1区。该研究首次报告了HCV NS5A区有来自E1区的漂移基因片段, 并对其临床和生物学意义进行了初步讨论。
The purpose of this study is on an inserting sequence element(IS element)in NS5A(non structure 5A)region of HCV genome and to explore its original source.A nucleotide segment from 6871 to 7121 in NS5A was amplified by using RT PCR method.The product sequencing was directly analysed and compared with HCV J genome(GenBank,D20908).A piece of nucleotide segment of 48bp was found,which inserted between 7041 to 7043 of HCV genome instead of the nucleotide GGC.The segment of this piece of nucleotide is same as the one from 941 to 989 in E1 region of HCV J genome,which the authors believe that is from.This is the first report of IS element from E1 region to NS5A in HCV genome.The clinic and biological significance was discussed in this paper as well.
