目的 :探讨bcl 2反义寡核苷酸诱导HL 6 0细胞的凋亡。方法 :将人工合成的bcl 2反义寡聚脱氧核苷酸片段与HL 6 0细胞共培养 ,在作用的第 0天、3天、5天通过免疫组化法 (APAAP法 )检测细胞内bcl 2蛋白表达水平的变化 ,通过流式细胞仪分析细胞凋亡的情况。结果 :bcl 2反义寡核苷酸可下调HL 6 0细胞内bcl 2的蛋白表达水平 ,其作用与其浓度和时间呈正相关 ,同时可诱导细胞凋亡 ,而bcl 2正义寡核苷酸及对照无此作用。结论 :bcl 2反义寡核苷酸诱导HL 6 0细胞凋亡的主要机制之一是下调bcl

Objective:To investigate the effect of bcl 2 antisense oligodeoxynucleotides on inducing HL 60 cells apoptosis. Methods:Synthesized bcl 2 antisense oligodeoxynucleotides were added to in vitro HL 60 culture system. We used immunocytochemical staining assay to detect the level of bcl 2 protein expression at day 0,3,5,and used the flow cytometer to detect the cellular DNA content to determine the cell apoptosis indirectly. Results:bcl 2 antisense oligodeoxynucleotides reduced the level of bcl 2 protein expression in HL 60 cells by the dose dependent manner and time dependent manner,and induced cells apoptosis.There was no difference on the effect of the sense oligodeoxynucleotides while compared with the control.Conclusion:It is one of the mainly mechanism by which bcl 2 antisense oligodeoxynucleotides induce HL 60 cells apoptosis to reduce the level of bcl 2 protein expression.
