摘要
目的 :探讨全反式维甲酸 (all transretinoicacid ,ATRA)诱导肿瘤细胞分化的分子机制。方法 :采用Southern和Northern杂交方法对本室用ATRA诱导人肺腺癌细胞GLC 82 ,经消减杂交所构建的cDNA消减文库进一步鉴定 ;序列分析后用RT PCR方法研究胎儿组织中的表达情况。结果 :分离到一为ATRA激活表达的基因—PIGB1,该基因与人PIG B (phosphatidylinositolglycanofcomplementationclassB)基因高度同源 ,在多种胎儿组织中均有表达。结论
Objective:To explore the molecular mechanism of cancer cell differentiation induced by all trans retinoic acid (ATRA ). Methods:Southern and Norhtern hybridization methods were used to screen the cDNA subtractive library constructed from ATRA induced human lung adenocarcinoma cell line GLC 82 in our laboratory. After sequence analysis,RT PCR method was used to analyze the expression pattern in human fetal tissues. Results:PIGB1,a gene highly homologous to phosphatidylinositol glycan of complementation class B (PIG B),up regulated by ATRA,was cloned. PIGB1 expresses in various human fetal tissues. Conclusion:PIGB1 may be involved in differentiation of lung adenocarcinoma cell induced by ATRA.
