平阳霉素诱导人食管癌细胞EC8712凋亡的研究 被引量:8

摘要 】目的 :以平阳霉素诱导食管癌细胞凋亡 ,并探索药物浓度与诱导凋亡之间的关系。材料与方法 :应用电镜、DNA电泳、TUNEL、荧光显微镜、激光共聚焦扫描显微镜等手段 ,观察食管癌细胞在不同浓度平阳霉素作用不同时间后的变化特征及诱导凋亡之作用。结果 :细胞形态学及DNA水平的变化均证实靶细胞发生了凋亡 ,但较大浓度较长时间的平阳霉素作用可致其坏死 ;平阳霉素对EC8712凋亡的诱导效率呈浓度、时间依赖性增强。结论 :一定浓度平阳霉素作用一定的时间可以诱导食管癌细胞株EC8712发生凋亡。诱导癌细胞凋亡可能是平阳霉素化疗作用的重要机制。临床化疗时可根据量效关系合理用药以诱导肿瘤细胞达到最大的凋亡率 ,而又不致其坏死。 Objectives:Pingyangmycin was used in vitro to induce apoptosis in human esophageal carcinoma (EC) cell line EC8712 and the concentration effect relationship was studied. Methods:Transmission electron microscopy (TEM), electrophoresis of DNA, terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL), fluorescence microscopy and confocal laser scanning microscopy (CLSM) were used to analyse the characteristic cellular changes. Results:Pingyangmycin used in proper concentration may induce apoptosis in EC8712, but higher concentration of the drug can lead necrosis of the cells. Pingyangmycin induced apoptosis of EC8712 cells is in a concentration dependent manner. Conclusions:Apoptosis of EC8712 can be induced by pingyangmycin within proper concentration after a long enough time. Apoptosis induction may be the major mechanism in chemotherapy of esophageal cancer with pingyangmycin. It is suggested that pingyangmycin should be used rationally according to the concentration effect relationship in clinical chemotherapy in order to induce the tumor cells to maximal apoptotic rate without necrosis and toxic or side effects.
