摘要
目的 观察热疗和顺铂对放射治疗的增强作用,以探索一种临床更为有效的肿瘤治疗方法。方法 采用X线照射前或后的人卵巢癌细胞A2780S( 对顺铂敏感) 和A2780CP( 抗顺铂) 用顺铂和热疗处理,观察两者对亚致死剂量辐射损伤修复(SLDR) 的抑制作用。结果 热疗(40℃,1 小时) 或顺铂处理1 小时均不能抑制这一修复,但两者联合应用则对两个细胞系均有明显效果。SLDR 的受抑程度取决于处理的时机。对于抗顺铂的A2780CP,可观察到SLDR的抑制,而在敏感细胞A2780S,除此之外还能观察到放疗敏感性的提高。结论 单独应用作用有限的热疗和低浓度顺铂联合应用可明显增强放疗疗效。
Objective To determine whether combination treatment with hyperthermia and cisplatin could inhibit repair of sublethal radiation damage in human ovarian cancer cells.Methods Human ovarian carcinoma cell lines A2780 S(sensitive to cisplatin) and A2780 CP(resistant to cisplatin) were used in this study.Cisplatin at concentrations of 1 or 3 μg/ml was given in combination with 1 hour of heating at 40℃ either before or after irradiation.The inhibition of cancer cell growing was determined using a colony forming assay.Results Neither hyperthermia nor cisplatin treatments alone affected sublethal radiation damage repair(SLDR).The combined treatment showed an effect in both cell lines and was treatment sequence dependent.The effect was greater in A2780 S.Conclusions The data show that combined treatment of cisplatin and hyperthermia may have clinical efficacy at cisplatin concentrations and hyperthermia temperatures which by themselves have little to no effect.\;
