摘要
目的 :探讨康莱特 (KLT)对肾癌细胞系 (GRC 1)的放射增敏作用和机制。材料和方法 :利用细胞克隆技术进行剂量 -存活曲线分析 ,末端脱氧核苷酰转移酶法检测细胞凋亡 ,免疫细胞化学法分析bcl-2和PCNA基因表达。结果 :0 2mg/mlKLT组D0 值为 90 44cGy ,空白对照组和空白乳组D0 值分别为 13 1 0 9cGy和 13 9 40cGy ,统计学处理P <0 0 5。 0 2mg/mlKLT处理GRC 1细胞后 ,M 1区阳性细胞为 89 76 % ,空白乳组为1 0 2 %。 0 2mg/mlKLT组GRC 1细胞bcl 2和PCNA基因表达的标记指数 (LI)分别为 6 6 0 %和 15 2 % ,空白乳组LI分别为 2 5 %和 0 % ,统计学处理差异显著 (P <0 0 1)。结论 :0 2mg/mlKLT具有明显提高GRC 1细胞放射敏感性的作用 ,其作用机制是诱发GRC 1细胞凋亡 ,抑制GRC 1细胞bcl
Objective:To investigate the influence of Kang Lai Te injection (KLT) on radiosonsitivity of renal carcinoma cell lines(GRC 1) after ionizing irradiation and its mechanism Materals and Methods:Exposed to lipase 10ul/ml and 0 2 mg/ml KLT respectively for 48h,the colony formation ability of GRC 1 cells after 10mv Xray irradiation was analysed by Linear quadratic formulation The apoptotic percentages of GRC 1 cell lines were measured by TUNEL The expression of bcl 2 and PCNA genes were detected by immunohistochemistry assay Results:The D 0 values of GRC 1 cell lines were 90 44 cGy,and those of control group and lipase group were 131 09 cGy and 139 40 cGy respectively(P< 0 05),there were no significant difference between the values of lipase and control group The apoptotic percentages of GRC 1 cell lines exposed to KLT and lipase group were 89 76%and 1 02%respectively The LI of KLT and lipase group were 6 60%and 25%respective (P< 0 01) Conclusion:KLT could increase radiosensitivity of GRC 1 cell lines,induce GRC 1 cell apoptosis,down regulate expression of bcl 2 gene and up regulate expression of PCNA
