摘要
目的研究1,25-二羟维生素D3[1,25(OH)2D3]对慢性阻塞性肺疾病(COPD)气道炎症的作用与机制。方法雄性Wistar大鼠30只,分为对照组(A组)、COPD组(B组)和1,25(OH):D3+CPPD组(C组),每组10只,用熏香烟加气管注内毒素法建立大鼠COPD模型,在C组腹腔注射1,25(OH)2D3,支气管肺泡灌洗液(BALF)细胞计数及分类,ELISA分析BALF中IL-8、TNF—α水平,流式细胞仪检测各组大鼠外周血清CD4、CD8T细胞亚群与肺组织树突状细胞的表达,HE染色观察气道炎症情况。结果B组与c组大鼠均出现气道炎症改变。但C组程度减轻。B组与c组大鼠血CD4、CD4/CD8较正常对照组降低,且BALF中细胞总数、中性粒细胞计数与TNF-α、IL-8的浓度升高,差异。均有统计学意义,其中c组表达较B组低,差异有统计学意义。结论1,25(OH)2D2减少COPD大鼠炎症细胞生成与炎症因子的表达,减轻气道炎症,改善免疫功能。
Objective To investigate the effect and mechanism of 1,25-Dihydroxy vitamin D3 (1,25(OH)2D3) on inflammation in chronic obstructive pulmonary disease (COPD). Methods Thirty male Wistar rats were randomly divided into three groups, A: control group, B.. COPD group, C: 1,25(OH)2D3 +COPD group. B and C groups was established by cigarette inhalation and LPS was instilled intratracheally once for every two weeks. 1,25 (OH)2 D3 was administered intraperitoneally in C group. Bronchoalveolar lavage fluid (BALF) was collected for total and differential cell counts,and the concentrations of IL-8 and tumor necrosis factor alpha (TNF-α) in BALF were detected by ELISA. The levels in serum of CD4,CD8 and the levels in lung of CDllc,CD80 to mark DC were measured by flow cytometer, pathologic changes of airways and lung tissues was observed by hematoxylin and eosin (HE). Results The pathologic changes of airways and lung tissue of B and C groups, but C group were signficanfly light than 13 group ( P d0.05). The levels of serum CD4 and CD4/CD8 were lower in B and C groups than that in the control group, and IL-8 and TNF-α inserum and BALF total and PMN cell counts were higher in B and C groups than in the control group ( P 〈0.05) ,and those were lower in C group than in B group ( P 〈0.05). Conclusions 1,25(OH)2D3 can relieve airway inflammation, diminish inflammatory cell and inflammatory factors in COPD rats, improve immunological function.
