小檗碱抑制人子宫内膜癌细胞增殖及转移机制的研究 被引量:8

摘要 目的探讨小檗碱抑制人子宫内膜癌Ishikawa细胞增殖和转移的机制。方法四甲基偶氮唑蓝法检测小檗碱对人子宫内膜癌Ishikawa细胞株的抑制作用;流式细胞仪观察对细胞周期分布的影响;ELISA法检测细胞培养上清液中炎性因子白细胞介素-8(IL-8)的浓度变化;应用Western印迹法检测经小檗碱干预该细胞48 h后,细胞核因子-кB(NF-кB)信号途径中p65的变化。结果小檗碱可显著抑制Ishikawa细胞生长,呈时间剂量依赖性。小檗碱可阻滞细胞于G0/G1期。细胞培养上清液中IL-8的浓度随着干预时间延长而减少(P<0.05)。小檗碱作用48 h后,细胞中NF-кB p65表达明显下降(P<0.05)。结论小檗碱具有抗人子宫内膜癌Ishikawa细胞增殖作用,将细胞阻滞于G0/G1期,抑制细胞产生IL-8,并能够阻断NF-кB信号通路,从而抑制子宫内膜癌的转移。 Objective To study the effect of Berberine on human endometrial carcinoma cell line Ishikawa.Methods The changes of cell proliferation of Ishikawa cells treated with Berberine were observed by using MTT assay.The cell cycle expressions were determined by flow cytometry.The concentration of IL-8 in the supenmatant fluid was measured by ELISA.And NF-кB p65 were determined by western blot assay.Results Berberine could inhibit the proliferation of Ishikawa cells in vitro significantly in a dose-and time-dependent manner(P〈0.05).The G0/G1 phase arrest was induced by treatment after 48 hours(P〈0.05).The concentration of IL-8 in the supenmatant fluid were decreased.Cellular NF-кB p65 levels decreased in Berberine-treated cells after 48 hours(P〈0.05).Conclusion Berberine could inhibit Ishikawa cell proliferation,induce G0/G1 phase arrest,and inhibit the metastasis of endometrial carcinoma by interfering with the NF-кB signaling pathway.
