GIT1发夹结构对成骨细胞内局部黏附激酶的磷酸化和分布的影响 被引量:1

Effect of GIT1 hairpin on phosphorylation and distribution of focal adhesion kinase in osteoblasts

摘要 目的探索G蛋白偶联受体激酶结合蛋白1的RNA发夹结构(GIT1-RNAh)对成骨细胞内局部黏附激酶(FAK)的磷酸化和分布的影响。方法成功构建含GFP-RNAh和GIT1-RNAh的腺病毒,在血小板衍生生长因子(PDGF)刺激的条件下,免疫荧光染色法检测成骨细胞内FAK的蛋白分布,Western blot检测FAK的磷酸化水平,Image J软件检测成骨细胞的扩增面积。结果与感染GFP-RNAh的成骨细胞相比,在PDGF刺激条件下,GIT1-RNAh明显抑制FAK磷酸化水平(P<0.05),扰乱FAK的分布,抑制成骨细胞的面积扩增(P<0.05)。结论 GIT1-RNAh通过扰乱FAK的分布及其磷酸化抑制成骨细胞的面积扩增。 Objective To investigate the effect of the RNA hairpin of G protein coupled receptor kinase interacting protein 1(GIT1-RNAh) on the phosphorylation and distribution of focal adhesion kinase(FAK) in osteoblasts.Methods The adenovirus containing GFP-RNAh or GIT1-RNAh was successfully constructed.After stimulated with platelet-drived growth factor(PDGF),the protein distribution of FAK,phosphorylation level of FAK in osteoblasts and the amplified area of osteoblasts were detected by immunofluorescence staining,Western blot and Image J software,respectively.Results Compared with the osteoblasts infected with GFP-RNAh,GIT1-RNAh significantly inhibited the phosphorylation level of FAK(P〈0.05),disturbed the distribution of FAK,and inhibited the amplified area of osteoblasts(P〈0.05) after PDGF stimulation.ConclusionGIT1-RNAh inhibits the amplified area of osteoblasts via disturbing the distribution and phosphory-lation of FAK.
