红景天苷诱导人胎盘间充质干细胞分化为肝细胞样细胞

Salidroside induces human placenta mesenchymal stem cellsto differentiate towards hepatocytes-like cell in vitro

目的探讨人胎盘间充质干细胞(PMSCs)在红景天苷作用下是否可以转化为肝细胞样细胞。方法分离培养人PMSCs,采用红景天苷和成纤维细胞生长因子4(FGF-4)联合应用对PMSCs进行诱导分化,倒置显微镜下每日观察诱导后细胞形态的变化;在诱导第7、14、21天,收获诱导细胞,免疫荧光染色检测CK18的表达,同时收集培养上清液,ELISA法检测白蛋白(ALB)浓度。结果在红景天苷和细胞因子的作用下,经过21 d的体外培养,PMSCs由长梭形变为多边形、类圆形,并具有肝细胞特异的表型和功能。免疫荧光可检测到诱导后细胞内CK18表达,培养上清液中有ALB的分泌。结论 PMSCs在红景天苷FGF-4作用下在体外能转化为肝细胞样细胞。

Objective To evaluate induction effect of salidroside to differentiate human placenta mesenchymal stem cells（PMSCs） towards hepatocytes-like cell in vitro.Methods PMSCs of passage 2 were incubated in differentiation DMEM medium （high glycose）,supplemented with 5%FBS,fibroblast growth factor（FGF）-4 10ng/ml and salidroside 10μM.Cell morphology was observed under inverted microscope every day after the induction.PMSCs were harvested on the 7^th,14^th and 21^st day for analyzing typical hepatic characteristics,such as cytokeratin（CK）18 and protein expression of ALB.Results With the induction of salidroside and CK18,PMSCs after 21 days incubation shaped to be rounded and irregular,which showed the characteristics similar to the hepatocytes of expressing CK18 in the cells（detected by immunofluorescence analysis） and ALB in the supernatant（examined by ELISA analysis）.Conclusion Salidroside combined with FGF-4 in vitro may induce human PMSCs to differentiate towards hepatocytes.
