脐血输注对再生障碍性贫血患者细胞因子的影响

Effects of cord blood transfusion on cytokines in serum and bone marrow in patients with aplastic anemia

摘要 目的探讨再生障碍性贫血(AA)患者脐血输注治疗前后细胞因子水平的变化。方法用ELISA法检测脐血输注前后20例AA患者骨髓及外周血中白细胞介素(IL)2、IL-3I、L-11、促血小板生成素(TPO)、干扰素γ(IFN-γ)水平变化。结果与输注前比较,脐血输注后AA患者外周血中IL-2、IFN-γ水平降低[(127.40±79.83)pg/ml vs.(90.77±4.86)pg/ml、(9.86±5.35)pg/ml vs.(4.48±3.35)pg/ml](P<0.05),IL-3水平升高[(99.13±55.31)pg/ml vs.(141.22±33.13)pg/ml)](P<0.05);脐血输注后,AA患者骨髓IL-2水平明显降低[(40.20±17.40)pg/ml vs.(5.87±0.96)pg/ml](P<0.05)。结论脐血输注可能通过改变造血环境中细胞因子的浓度或比例改善AA患者的造血功能。 Objective To investigate the effects of cord blood transfusion on cytokines in serum and bone marrow(BM) in patients with aplastic anemia(AA).Methods The levels of IL-2,IL-3,IL-11,TPO and IFN-γ in serum and BM were measured by ELISA before and after cord blood transfusion in 20 AA patients.Results Compared to before,the levels of IL-2 and IFN-γ in serum were significantly decreased[(127.40±79.83)pg/ml vs.(90.77±4.86)pg/ml,and(9.86±5.35)pg/ml vs.(4.48±3.35)pg/ml],the level of IL-3 in serum was increased[(99.13±55.31)pg/ml vs.(141.22±33.13)pg/ml)],and the level of IL-2 in BM was significantly decreased [(40.20±17.40)pg/ml vs.(5.87±0.96)pg/ml] after cord blood transfusion(P〈0.05).Conclusion Cord blood transfusion can improve hematopoiesis by changing the concentration or proportion of cytokines in microenvironment.
