摘要 目的初步探索超声联合造影剂SonoVue辐照PC3和LNcap细胞的适宜条件,并比较超声辐照对两种细胞膜通透性和存活率的影响。方法设不同的声强、辐照时间和造影剂浓度,采用1 MHz、连续声波于37℃水浴中分别辐照PC3和LNcap细胞。以流式细胞仪测量细胞发光率,以台盼蓝拒染法测算细胞存活率。结果两种细胞的适宜辐照条件均为声强0.9 W/cm2、辐照时间40 s、造影剂浓度20%。辐照声强为1.5 W/cm2、辐照时间20 s、SonoVue浓度为20%时,PC3细胞的发光率高于LNcap细胞;在其余辐照条件下,PC3细胞的发光率小于或者等于LNcap细胞。PC3细胞的存活率在所有的辐照条件下均高于LNcap细胞(P<0.05)。结论优化各项辐照参数可以安全有效地增加细胞膜的通透性。 Objective To investigate the optimal ultrasonic irradiation conditions of PC3 and LNcap cells,and to compare the effect of ultrasound irradiation on the cell membrane permeability and cell survival rate between the two types of human prostate cancer cells.Methods PC3 and LNcap cells were exposed to 1 MHz,continuous waves in 37℃ water bath,respectively.Different parameters such as ultrasound intensity,exposure duration and microbubble concentration were applied.The cell luminosity factor was detected by flow cytometry,and the cell survival rate was calculated with trypan blue exclusion.Results The optimal parameters for both types of cells were intensity 0.9 W/cm2,exposure duration 40 s,contrast agent concentration 20%.When exposed to ultrasound intensity of 1.5 W/cm2 for 20 s in the presence of 20% contrast agent,the luminosity factor of PC3 cells was higher than that of LNcap cells.The luminosity factor of PC3 cells was lower than or the same as that of LNcap cells when exposed to other parameters.On conditions of different parameters,the survival rate of PC3 cells was always higher than that of LNcap cells(P0.05).Conclusion Optimizing the exposure parameters can safely and efficiently increase the cell membrane permeability.
