吲哚青绿联合光照对兔晶状体上皮细胞影响的实验研究 被引量:4

Effects of indocyanine green combined with light on lens epithelial cells in rabbits

摘要 目的观察兔晶状体囊外摘出术(extracapsular lens extraction,ECLE)中使用吲哚青绿(indocyanine green,ICG)水分离联合光照对残留晶状体上皮细胞(lens epithelial cells,LEC)的影响。方法 40只兔随机分成ICG+光照组、ICG组、光照组和空白对照组,每组各10只兔(20眼)。各组双眼均行ECLE术。ICG+光照组用5g·L-1ICG做水分离联合卤素灯光源照射;ICG组用5g·L-1ICG水分离;光照组用BSS水分离联合卤素灯光源照射;空白对照组用BSS做水分离。术后第1天、第7天时,每组随机处死5只兔。TUNEL方法检测各组术后LEC的凋亡并计算各组的TUNEL阳性细胞率(percentage of TUNEL positive cells,PT)。结果术后第1天,ICG+光照组LEC呈单层排列,其他各组LEC呈单层或复层排列;术后第7天,ICG+光照组LEC少或无,其他3组LEC呈多层束状或成簇排列。术后第1天,ICG+光照组PT值为(42.30±7.60)%,高于其他3组,ICG组、空白对照组均为0,光照组PT值为(0.13±0.02)%,差异均有显著统计学意义(均为P<0.0001),ICG组、光照组及空白对照组PT值组间差异无统计学意义(P>0.05)。术后第7天,ICG+光照组囊膜上无或仅见少数LEC生长,未求PT值,ICG组、光照组及空白对照组PT值分别为(3.70±1.64)%、(3.30±1.36)%、(3.80±0.79)%,组间差异均无统计学意义(均为P>0.05)。结论兔ECLE术中采用5g·L-1ICG水分离联合光照,可致术后早期LEC大量减少,LEC凋亡参与了这一过程。 Objective To investigate the effects of indocyanine green(ICG) hydrodissection combined with visible light on residuary lens epithelial cells(LEC) during extracapsular lens extraction(ECLE) of rabbits in vivo.Methods Forty rabbits were randomly divided into ICG+light group,ICG group,light group and control group,10 cases(20 eyes) in each group.ECLE were performed in rabbit dual eyes of each group.When performing hydrodissection,5 g·L-1 ICG combined with light,5 g·L-1 ICG,BSS combined with light and BSS were used in ICG+light group,ICG group,light group and control group,respectively.Five rabbits were sacrificed randomly at postoperative 1 day and 7 days,respectively.LEC apoptosis was detected by TUNEL method,and percentages of TUNEL positive cells(PT) were calculated for each group.Results At postoperative 1 day,LEC were aligned in single layer in ICG+light group,while in single or multi-layer in other groups.At postoperative 7 days,no or only few LEC were found in ICG+light group,while LEC became multi-layer with fusiform in other groups.At postoperative 1 day,PT were(42.30±7.60)%,0,(0.13±0.02)%,0 in ICG+light group,ICG group,light group and control group,respectively,ICG+light group was higher than other groups,there were significant differences(all P0.000 1),while there was no statistical difference among other groups(P0.05).At postoperative 7 days,PT was not calculated in ICG+light group because no or only few LEC were observed on capsules,PT in ICG group,light group and control group were(3.70±1.64)%,(3.30±1.36)% and(3.80±0.79)%,respectively,there was no statistical difference among groups(all P0.05).Conclusion ICG(5 g·L-1) hydrodissection combined with visible light in experimental ECLE of rabbit decreases the number of LEC in early postoperative stage,the apoptosis of LEC may be take part in the process.
