紫外线减毒弓形虫ZS1株滋养体免疫小鼠的体液免疫反应 被引量:2

摘要 探讨紫外线减毒弓形虫活疫苗在小鼠体内的免疫保护性和体液免疫反应。采用波长为2 537A 的紫外灯照射弓形虫ZS1 株滋养体,照射距离5cm ,照射时间分别为30 、60 、90min 和120min 。小鼠分为4 组,组1 为免疫组,组2 为免疫攻击组,组3 为感染组,组4 为正常对照组。接种照射30min 虫体的小鼠在接种后7 ~8d 全部死亡,其他免疫接种小鼠均正常存活,各组织未查见滋养体、假包囊或包囊。用同株滋养体攻击感染,免疫攻击组小鼠较单感染组小鼠存活时间延长。经ELISA及IHA检测,免疫组及免疫攻击组血清IgG抗体滴度增高,提示紫外线减毒弓形虫ZS1 株疫苗免疫小鼠后能够刺激机体产生一定的免疫保护力,其中体液免疫可能发挥着一定作用。 To explorethe protective effectand the humoralim munereaction of UV attenuated ZS1 strain of Toxoplasma gondiiin mice.The ZS1 strain trophozoites of T.gondii were irradiated by UV light with 2 537 A wavelength , 5cm in distance and withirradiatedtime30 ,60 ,90min and 120min respectively. Mice were dividedinto 4 groups.Group 1 vaccinated alone,group 2 challenged with normal ZS1 trophozoites at 45th day after vaccination ,group 3infected alone,and group 4 was normalcontrol. The mice inoculated with the trophozoitesirradiated 30 minutes were alldead at7 ~8d.Othervaccinated micesurvived normally,and notrophozoite,pseudo cystorcyst wasfound in theirtissues. Mice ofgroup 2 survivedlongerthan thosein group 3 .The value ofIgGin sera detected by ELISAand IHAin responsetosoluble Agof T.gondii wassignificantlyenhancedin both group 1 and group 2 .Ithasbeensuggestedthatthe UV attenuated vaccine of T.gondii of ZS1 strain could induce certain protective im munity in mice,in which humoralim mune may play a role.
