摘要
目的 研究胎儿酒精中毒脑损伤的细胞分子机制.方法 腹腔注射中毒剂量酒精至生后3~10dSD大鼠,制作急性酒精中毒模型;6小时后提取海马细胞DNA,并用琼脂糖凝胶电泳分离DNA;同时制作海马结构的石蜡切片,尼氏染色观察海马及齿状回细胞的结构.结果 酒精注射组海马细胞DNA断裂,断裂片段集中在200bp、400bp、900bp等;海马CAI区,齿状回颗粒层、多形层的细胞数均明显减少;CAl区锥体细胞胞核染色体明显浓缩呈团块状并聚集在核膜周围;颗粒细胞层胶质细胞增生;多形层细胞肿胀,细胞核染色体消失.结论 酒精破坏发育期大鼠海马结构,引起海马细胞凋亡或坏死.确切机制有待进一步阐明.
objective To study the molecular and cellular mechanism of the brain injury in fetal alcohol syndrom. Methods Intrapri-toneal injection of 2000 proof ethanol(2.5 g.kg)to P3 ~ P10 SD rats were performed.The molecular and morphology experiments were started six hours later. Apoptotic DNA laddering kit-Ethidium Bromide was used to detect DNA from the hippocampi of the rats. Nissl stain was used to observe the cytological morphology of paraffin-embedded hippocampal tissues. Result Ethanol induced the nuclear DNA fragmentation of hip-pocampal cells, there were 200bp,400bp,900bp or over DNA fragments.The most one was the 400bp fragment. Ethanol diminished the cells in CA1 pyramidal layers of Ammon's horn,granular layers and polymorphological cell layers(CA4)of dentate gyrus.The chromatin condensations occurred in pyramidal nuclei in CA1 area. There were glial cell proliferation in granular layer. Some cell swollened and the chromatin disap-peared in the polymorphological layers. Conclusion Ethanol can destroy the molecular and cytological structure of the developing hippocam-pus. There are either apoptosis or necrosis.We discuss the potential mechanism in the article.
