摘要
本项目研究从制样方法、色谱柱筛选、检测波长确认、定容的试剂确认以及方法的重复性、再现性与准确度等方面全面考量与优化液相色谱检测草酸的分析方法,摸索一套快速、准确可行的草酸含量的检验方法;建立不同果蔬基质、不同草酸含量范围的液相色谱检测线性回归方程;浓度范围为1~500μg/mL,回归方程为y=25.55x+0.139,线性相关性R^2=0.9998良好,最低检出限5(S/N)为0.1μg/mL。精密度误差RSD<5%,回收率98.0%~105.4%,并在5家实验室进行验证,实现立项时的研究目标。
This project studies comprehensively consider and optimize liquid chromatography analysis of oxalic acid from sample preparation, column selection, eomfirnnation of detection wavelength confirmation of constant volume of the reagent and methods repeatability, reproducibility and accuracy.Establish the linear regression equation of liquid chromatography detection of the content range of different fruits and vegetables and different oxalic acid: concentration range 1-500μg/mL: linear regression equation y.=25.55X+0.139, linear correlation coefficient R2=0.9998. RSD〈5%.the lowest detection limit of 0.1μg/ mL, the recovery of 98.0%-105.4%.and verity methods of 5 laboratories, achieved project objectives of research.
