摘要
目的观察首乌延寿丹对兔骨关节炎(Osteoarthritis,OA)的防治作用。方法将24只日本大耳白兔随机分成空白对照组、模型组、首乌延寿丹组。ACLT术[1]制作OA模型。于第12周末观察软骨病理形态学改变;免疫组化法测定软骨Ⅱ型胶原及基质金属蛋白酶-1(MMP-1)的表达。结果以ACLT术成功建立了OA模型。首乌延寿丹组病变明显减轻,Ⅱ型胶原的蛋白表达明显升高(P<0.01);MMP-1表达明显降低(P<0.01)。结论首乌延寿丹可明显抑制OA发展,这一作用可能是通过增加Ⅱ型胶原的蛋白表达、下调MMP-1表达来实现的。
【Objective】 To investigate the preventive effects of SWYSD on the rabbits with Osteoarthritis(OA).【Methods】 24 Japan big-ear white rabbits were randomly divided into normal control group﹑model group﹑SWYSD group.The OA model was established with ACLT.Pathological changes of collagen was observed and the expression of typeⅡcollagen and metalloproteinase-1(MMP-1) was detected by immunohistochemistry in the 12th week.【Results】 The OA model was successfully established with ACLT.There was severe pathological lesion in model group;The expression of typeⅡcollagen in model group were upwnregulated(P0.01),the expression of MMP-1 in model group were downregulated(P0.01).【Conclusion】 SWYSD can prevent the formation of OA by upregulating the expression of typeⅡcollagen and downregulating the expression of MMP-1.
