摘要 目的建立不稳定斑块动物模型,观察瑞舒伐他汀是否通过影响基质金属蛋白酶(matrix metallo-proteinase,MMP)-2,9的表达而稳定动脉粥样硬化不稳定斑块。方法健康雄性新西兰大白兔30只,随机分为空白对照组(对照组,n=10)、不稳定斑块组(VAS组,n=10)和瑞舒伐他汀组(RSV组,n=10,1mg/kg/d)。空白对照组给予正常饮食。VAS组及RSV组通过高脂饲料+球囊扩张拉伤术建立不稳定斑块动物模型后,RSV组再给予瑞舒伐他汀干预治疗1个月。处死全部大白兔,HE切片测算腹主动脉血管内膜/中膜(I/M);免疫组化分析MMP-2,9表达。结果 HE切片I/M比值示,VAS组较对照组明显升高,RSV组较VAS组显著降低,P<0.001有统计学意义。免疫组化示,RSV组MMP-2,9的表达较VAS组显著减少,P<0.05有统计学意义。结论瑞舒伐他汀通过抑制MMP-2,9的表达来减轻动脉粥样硬化不稳定斑块的形成。 【Objective】 Established animal model of vulnerable plaque to observe whether rosuvastatin could stabilize vulnerable plaque by influencing the expression of matrix metalloproteinase(matrix metalloproteinase,MMP)-2,9.【Methods】 Thirty healthy male New Zealand white rabbits were randomly divided into three groups:control group(n =10),vulnerable plaque group(n=10) and rosuvastatin group(n=10,1mg/kg/d).Animals in control group were fed on a regular diet,the others received hyper-lipid diet+balloon-injury of abdominal aortic surgery to establish animal model of vulnerable plaque.After that,RSV group received hyper-lipid diet with rosuvastatin for one month,respectively.Then killed all.Measured and calculated the abdominal atherosclerotic vascular HE sections intima to media(I / M).Immunohistochemical staining of MMP-2,9 antigen expression was performed.【Results】 HE sections I/ M ratio in VAS group were higher than that in Control group,I/ M ratio in RSV group were lower than that in VAS group.P0.001 was statistically significant.Immunohistochemistry showed that,MMP-2,9,P0.05 was statistically significant.【Conclusion】 Rosuvastatin might reduce the formation of vulnerable plaque of abdominal atherosclerosis by inbiting the expression of matrix metalloproteinase-2,9.
