竹节参齐墩果酸型皂苷的乙醇提取工艺研究

摘要 目的确定竹节参齐墩果酸型皂苷的最佳乙醇溶液回流提取工艺条件。方法采用正交设计L9(34)方法,考察乙醇浓度、浸提时间、浸提温度和固液比等因素对提取率的影响。用分光光度法测定含量,并与水溶液提取法、超声提取法进行了对比。结果所考察的因素对竹节参齐墩果酸型皂苷提取的影响程度为:乙醇浓度>浸提温度>固液比>浸提时间,超声提取的最佳条件为:D2A2C2B1。结论与水溶液提取法、超声提取法进行了对比,乙醇溶液回流提取具有提取时间短、杂质少、收率高、实验条件易于控制,技术成熟等优点。 【Objective】 To optimize the ethanol extraction condition for oleanolic acid saponins in Panax japonicus C.A.Mey.【Methods】 According to the orthogonal design L9(34),the optimum ethanol extraction technique was sought through experimental investigation,and was compared with the aqueous extraction technics and ultrasonic extraction technics.The contents of oleanoclic acid were determined by UV-spectrophotometry.【Results】 The order of factors to affect the oleanolic acid extraction was first ethanol concentration,second refluxing temperature,third the ratio of solid to liquid,finally the refluxing time.The optimum ethanol extractions were A2B3D3C2.【Conclusion】 Compared with the aqueous extraction technics and ultrasonic extraction technics,refluxing with ethanol extraction time is short,less impurities,high yield,easy control of experimental conditions,mature technology and so on.
