摘要
目的观察五肽胃泌素(PG)及其受体拮抗剂丙谷胺(PGL)对人结肠癌细胞株HT-29表皮生长因子(EGF)和血管内皮生长因子(VEGF)表达的影响。方法体外培养结肠癌细胞株HT-29,用PG及PGL干预。放射免疫法测定EGF,ELISA法测定VEGF。结果在PG浓度为10^(-5)mol/L、10^(-6)mol/L时,PG组EGF明显高于空白对照组(P<0.01),PG+PGL组EGF明显低于PG组(P<0.01);在PG浓度为10^(-7)mol/L时,PG组与空白对照组以及PG+PGL组与PG组EGF含量比较差异均无统计学意义(P>0.05)。在PG浓度为10^(-5)mol/L时,PG组VEGF明显高于空白对照组(P<0.01),PG+PGL组VEGF明显低于PG组(P<0.01);在PG浓度为10^(-6)mol/L、10^(-7)mol/L,PG组与空白对照组以及PG+PGL组与PG组VEGF含量比较差异均无统计学意义(P>0.05)。结论五肽胃泌素能促进体外培养结肠癌细胞株HT-29中EGF和VEGF的表达,而其受体拮抗剂丙谷胺能部分阻断这一作用。提示胃泌素的促结肠癌细胞生长作用可能与促进EGF和VEGF表达有关。丙谷胺可部分抑制其作用。
Objective To investigate the effects of PG and its receptor antagonist PGL on secretion of EGF and VEGF in HT-29 cell lines in vitro.Methods The colonic cancer cell lines HT-29 was cultured in vitro and interfered with PG and PGL.The expression of EGF was detected by radioimmunoassay.The expression of VEGF was detected by ELISA.Results At the concentration of 10^(-5) mol/L and 10^(-6) mol/L,the content of the EGF in PG group was obviously higher than that in control group(P 0.01),and PG plus PGL group was less than that in PG group(P 0.01).At the concentration of 10^(-7) mol/ L,there was no difference between PG group and control group,similar result in comparison of PG plus PGL group and PG group(P 0.05).At the concentration of 10^(-5) mol/L,the content of VEGF in PG group was higher than that in control group(P 0.01),and PG plus PGL group was less than that in PG group(P 0.01).At the concentration of 10^(-6)mol/L and 10^(-7) mol/L,there was no difference between PG group and control group,similar result in comparison of PG plus PGL group and PG group(P 0.05).Conclusion It demonstrated that the PG could promote the secretion of EGF and VEGF in colonic cancer cell lines HT-29 in vitro,while PGL may inhibit its effects.Gastrin could promote the growth of colonic cancer maybe caused by stimulating the secretion of EGF and VEGF,while the PGL could inhibit partly.
