摘要
目的:评价特异性钠泵抑制剂哇巴因在体外对非小细胞肺癌A549细胞增殖与死亡的影响。方法:利用细胞培养技术,采用MTT法、流式细胞技术、TUNEL法及电镜技术分别评价不同浓度哇巴因对A549增殖与死亡的影响。结果:当哇巴因浓度<1.8nmol/L时,可通过干扰细胞周期,显示出促进肿瘤细胞增殖作用。哇巴因浓度≥1.8nmol/L时,肿瘤细胞增殖受抑。哇巴因对A549细胞增殖的抑制作用呈明显的剂量依赖性和时间依赖性。高浓度哇巴因(≥100nmol/L)所致的A549细胞死亡以非凋亡性死亡为主。结论:哇巴因在适当浓度具有抗肺肿瘤活性,可能成为潜在的抗肺肿瘤药物。
Objective:To evaluate the effect of specific sodium pump inhibitor ouabain on the proliferation and death of A549 cells in vitro.Methods: Using MTT,flow cytomertry,TUNEL and electron microscopy to evaluate different concentration of ouabain on the proliferation and death of cultured A549 cells.Results: When concentration of ouabain was 1.8nmol/L,it stimulated the growth of cancer cells through interference of cells cycle.When concentration of ouabain was≥1.8nmol/L,proliferation of cancer cells was inhibited in time-dependent and dose-dependent way.When concentration was ≥100nmol/L,ouabain induced a non-apoptotic-related cells death process.Conclusion: Ouabain has anti-lung cancer effects in proper concentration,and may be a potential anticancer drug.
