调营饮对化疗所致骨髓抑制小鼠造血细胞因子的影响 被引量:3

摘要 目的观察调营饮对化疗所致骨髓抑制小鼠造血细胞因子的影响。方法将168只清洁级昆明小鼠,随机分为正常组、模型组、调营饮组、调营饮拆方组、六味地黄汤组、八珍汤组、当归补血汤组,每组24只。正常组予0.9%氯化钠注射液0.2 mL腹腔注射,其余各组予注射用环磷酰胺(CTX)100 mg/kg腹腔注射,每日1次,连续3 d。实验第4 d,调营饮组、调营饮拆方组、六味地黄汤组、八珍汤组、当归补血汤组分别给予相应药液0.2 mL灌胃,正常组、模型组予等容积蒸馏水灌胃,每日1次,连续10 d。分别于实验第4、7、10 d取小鼠眼球血,测外周血白细胞计数(WBC)、红细胞计数(RBC)。同时,第7 d采用放射免疫分析法检测血液中促红细胞生成素(EPO),之后取各组小鼠右侧股骨,采用免疫组化法观察粒-巨噬细胞集落刺激因子(GM-CSF)平均光密度值。结果实验第4、7 d模型组及各药物组WBC均低于正常组(P<0.05)。实验第10 d,模型组、调营饮拆方组、六味地黄汤组及当归补血汤组WBC均低于正常组(P<0.05)。模型组各时间RBC均低于正常组(P<0.05)。实验第4 d,调营饮组、调营饮拆方组及八珍汤组WBC较模型组升高(P<0.05);调营饮拆方组、六味地黄汤组RBC与模型组比较差异有统计学意义(P<0.05)。实验第7 d,调营饮组、八珍汤组WBC与模型组比较均明显升高(P<0.05);调营饮组、调营饮拆方组RBC高于模型组(P<0.05)。实验第10 d,各用药组WBC、RBC与模型组比较差异均有统计学意义(P<0.05),且调营饮组WBC高于调营饮拆方组、六味地黄汤组和当归补血汤组(P<0.05),调营饮组RBC高于其他各用药组(P<0.05)。模型组EPO高于正常组(P<0.05),GM-CSF平均光密度值低于正常组(P<0.05)。各用药组EPO、GM-CSF平均光密度值与模型组比较均有统计学意义(P<0.05),且调营饮组EPO、GM-CSF平均光密度值均高于其他各药物组(P<0.05)。结论调营饮可明显提高化疗致骨髓抑制小鼠EPO分泌及GM-CSF的表达,从而改善外周血细胞状况,促进骨髓造血功能恢复。 Objective To investigate the effect of Tiaoyangyin on hematopoietic cytokines in mice of chemotherapy-induced bone marrow suppression.Methods 168 mice were randomly seven groups,control group,model group,Tiaoyangyin group,Tiaoyangyin Decomposed Recipes group,Rehmanniae Decoction of Six Ingredients group,Decoction of Eight Ingredients group and Angelicae Sinensis Decoction for Supplementing Blood group.There were 24 mice in each group.Control group received intraperitoneal injection of 0.9% sodium chloride injection 0.2 mL,the rest of the group received intraperitoneal injection of cyclophosphamide(CTX) 100 mg/kg,once a day,continuous 3 days.Experiment No.4 d,Tiaoyangyin group,Tiaoyangyin Decomposed Recipes group,Rehmanniae Decoction of Six Ingredients group,Decoction of Eight Ingredients group and Angelicae Sinensis Decoction for Supplementing Blood group were given 0.2 mL of corresponding liquid fed,the normal group and model group were given equal volume of distilled water,once a day,continuous 10 d.White blood cell count(WBC) and red blood cell count(RBC) in peripheral blood from eye of mice blood at experiment No.4,7 and 10 d.while blood erythropoietin(EPO) were detected by radioimmunoassay at the first 7 d.Mean optical density value of GM-CSF from the right femur of mice were observed using immunohistochemical staining.Results WBC in model group and each treatment group was lower than that in control group at experiment No.4 and 7 d(P0.05).WBC in model group,Tiaoyangyin Decomposed Recipes group,Rehmanniae Decoction of Six Ingredients group and Angelicae Sinensis Decoction for Supplementing Blood group was lower than that in control group at experiment No.10 d(P0.05).There was no significant difference between Tiaoyangyin group,Decoction of Eight Ingredients group and control group on WBC(P0.05).RBC in model group was lower than that in control group at difference time(P0.05).WBC in Tiaoyangyin group,Tiaoyangyin Decomposed Recipes group and Decoction of Eight Ingredients group was higher than that in control group at experiment No.4 d(P0.05).There was significant difference between Tiaoyangyin Decomposed Recipes group,Rehmanniae Decoction of Six Ingredients group and model group on RBC(P0.05).WBC in Tiaoyangyin group and Decoction of Eight Ingredients group was significantly increased in comparison with that in model group at experiment No.7 d(P0.05).There were significant differences between each treatment group and model group on WBC and RBC at experiment No.10 d(P0.05).There were significant differences between each treatment group and model group on EPO and mean optical density value of GM-CSF(P0.05).Conclusion Tiaoyangyin can significantly increase GM-CSF secretion of EPO expression in mice of chemotherapy-induced bone marrow suppression,thereby improving the status of peripheral blood cells and promote functional recovery of bone marrow.
