血府逐瘀汤诱导内皮细胞迁移的机制研究 被引量:14

摘要 目的:探讨血府逐瘀汤促进内皮细胞迁移作用及其机制。方法:运用血清药理学方法,以1.25%,2.5%,5%的血府逐瘀汤含药血清和空白血清培养内皮细胞ECV304,采用划痕损伤法和侵袭实验检测药物对细胞迁移作用的影响,并分别通过酶联免疫法和硝酸还原酶法检测细胞培养上清液中血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)和一氧化氮(NO)浓度,荧光探针法检测胞内NO浓度,eNOS酶活性以及RT-PCR法检测VEGF,bFGF,eNOS基因的表达,以进一步阐明药物的作用机制。结果:1.25%含药血清可促进细胞在划痕平面上的迁移作用,5.0%含药血清可显著提高细胞侵袭穿越的迁移作用,而2.5%含药血清对这两类迁移均有促进作用。1.25%,2.5%,5.0%含药血清均可明显提高上清液中bFGF、胞内外NO浓度和胞内eNOS活性,只有2.5%含药血清能提高上清液中VEGF的含量。RT-PCR结果显示,1.25%,2.5%,5.0%含药血清均可提高bFGF的转录;2.5%和5.0%含药血清可上调eNOS的表达;而药物对VEGF的影响较复杂,仅2.5%含药血清上调VEGF的表达,1.25%含药血清却呈抑制作用。结论:血府逐瘀汤通过上调VEGF,bFGF,NO的表达与分泌,诱导内皮细胞迁移,从而发挥促血管新生功能。 Objective: To investigate the effect and mechanism of Xuefu Zhuyu Decoction inducing endothelial cells migration.Method: Serum pharmacology technique was adopted.Endothelial cells ECV304 were incubated with blank serum or Xuefu Zhuyu Decoction containing serum(XFZYD-CS) at final concentrations of 1.25%,2.5% and 5%.Cell migration was assessed by wound healing assay.Vascular endothelial growth factor(VEGF),basic fibroblast growth factor(bFGF) and nitric oxide(NO) level in the culture supernate were evaluated by ELISA and nitrate reductase method respectively;intracellular NO was studied by fluorescent probe;endothelial nitric oxide synthase(eNOS) activity was assessed and VEGF,bFGF and eNOS expression were evaluated by RT-PCR.Result: 1.25%XFZYD-CS could induce cell migration in the wound healing model,5%XFZYD-CS could induce cell migration in Transwell,and 2.5%XFZYD-CS could work both methods.All 3 concentrations of XFZYD-CS could elevate bFGF,NO level in the culture supernate,intracellular NO level and eNOS activity.Only 2.5%XFZYD-CS could elevate supernate VEGF level.RT-PCR results showed that all 3 concentrations of XFZYD-CS could upregulate bFGF expression;2.5% and 5%XFZYD-CS upregulated eNOS transcription;only 2.5%XFZYD-CS elevated VEGF expression,but 1.25%XFZYD-CS was opposite work.Conclusion: Xuefu Zhuyu Decoction could induce endothelial cell migration into angiogenesis via upregulating VEGF,bFGF and eNOS/NO pathways.
