二苯乙烯苷对H_2O_2诱导PC12细胞损伤的抗氧化作用 被引量:3

摘要 目的探讨2,35,4,'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(TSG)对H2O2诱导PC12细胞损伤的抗氧化作用。方法本实验分为正常对照组、H2O2处理组和TSG(0.11、1、0和50μmol/L)预处理组。采用MTT比色法检测细胞活性,活性氧探针检测细胞内活性氧(ROS)水平,用超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)试剂盒测定细胞的氧化应激变化。结果与正常对照组相比,细胞经H2O2诱导处理后,细胞存活率降低,胞内ROS水平增高,抗氧化酶活力降低,脂质过氧化产物MDA含量增加。不同浓度的TSG预处理后能减轻H2O2所致的细胞损伤,减少胞内ROS累积,增加细胞的抗氧化酶活力,减轻脂质过氧化反应。结论 TSG可抑制H2O2诱导的PC12细胞损伤,其机制可能与其抑制氧化应激有关。 Objective To observe the anti-oxidative effects of 2,3,5,4'-tetrahydroxystibene-2-O-β-D-glucoside(TSG) on H2O2-induced PC12 cell injury.Methods The PC12 cells were divided into normal control,H2O2-induced and TSG pre-treatment groups with different concentrations.Colorimetric assay method of MTT was used to measure the cell viability.Reactive oxygen species-sensitive fluorescent probe was employed to evaluate the levels of intracellular ROS.The assay kits of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and malondialdehyde(MDA) were used to detect the alterations of oxidative stress.Results Compared with the normal control,the cell viability and the activities of antioxidase were decreased,the intracellular ROS levels and MDA contents were increased in H2O2-induced group.H2O2-induced cell injury was relieved,the intracellular ROS accumulation and lipid peroxidation were decreased,the activities of antioxidase was increased in TSG pretreatment group.Conclusions TSG could inhibit the cell injury induced by H2O2 in PC12 cells,which might correlate with the anti-oxidant activity of TSG.
