摘要
以大麦麦麸为原料,对不溶性草酸氧化酶进行了部分纯化,比较了酶法与滴定法测定草酸的方法。结果表明,粗酶经酸热变性、淀粉酶水解、NaCl洗涤、去离子水洗涤4个步骤的纯化后,不溶性草酸氧化酶的纯化倍数为1.51;酶法与滴定法测定草酸无显著性差异。该技术有望用于草酸检测试剂盒的开发。
The partial purification of unsoluble oxalate oxidase from barley bran was explored.And the colorimetric method using unsoluble oxidase and the titration method for determining the oxalic acid content was compared.The results showed that the enzyme was purified 1.51-fold after sequential thermal and acid treatment,hydrolyzing with amylase,washing with NaCl and distilled water.No significant difference was observed between the colorimetric method and the titration method of determining wheat oxalic acid.This technology could be used in the manufacturing of oxalate detection kit.
