摘要
目的探讨丹参注射液联合骨髓间充质干细胞移植对新生大鼠缺氧缺血性脑损伤(HIBD)后海马神经元Bax、Bcl-2蛋白表达的影响及其治疗作用。方法建立新生SD大鼠HIBD模型,每组实验随机分为4组:假手术组(S组)、生理盐水对照组(C组)、MSCs治疗组(M组)和丹参+MSCs治疗组(DM组)。观察缺氧缺血组28d内生存情况;检测缺氧缺血前2h,缺氧缺血后12h、24 h,3、7、14、28d各时间点新生大鼠海马CA1区神经元Bax、Bcl-2蛋白的表达。移植后28d进行大鼠迷宫行为学实验。结果 DM组较C组和M组比较,Bax表达明显下调(<0.05),而Bcl-2表达显著上调(<0.05),生长发育和生存情况明显好于后两者。迷宫实验显示结论:丹参合并MSCs改善了新生大鼠HIBD后的生长发育,提高了其空间记忆能力。结论丹参合并骨髓间充质干细胞移植能减少Bax的产生,发挥对新生鼠HIBD时的脑保护和治疗作用。
Objective To study the effect of Danshen with intraperitoneal transplantation of mesenchymal stem cells(MSCs) on Bax and Bcl-2 level in neonatal rats with hypoxic-ischemic brain damage and explore the therapeutic effect of them.Methods Establish a model of hypoxic-ischemic brain damage in neonatal SD rats.SD rats were sham-operated and treated with saline injection(group S) or hypoxia-ischemia and treated with either saline(group C) or Danshen(group D) or Danshen + MSCs(group DM) by intraperitoneal injection.The rats were sacrificed after HIBD and the expression of Bax and Bcl-2 were assessed.Water maze was used to test rats' learning ability.Results Expression of Bax or Bcl-2 was lower or higher in group DM than that of group C.Behavior test showed that the escape latency in group DM with HIDB was shorter than those in group C and group D.Conclusions Danshen injection with and MSCs intraperitoneal could prevents apoptosis of neurous and promote brain repair and functional recovery.
