摘要 目的:探讨E-cadherin在结直肠癌中的表达及临床病理意义。方法:采用EliVisionTM plus免疫组织化学和原位杂交方法检测E-cadherin蛋白和mRNA在结直肠癌、结直肠腺瘤和结直肠癌旁正常黏膜组织中的表达。结果:E-cadherin蛋白和mRNA在结直肠癌中的阳性率显著低于结直肠腺瘤和结直肠癌旁正常黏膜组织;E-cadherin的异常表达与结直肠癌组织的分化程度、TNM分期、浸润深度及淋巴结转移相关,而与年龄、性别及肿瘤大小无关。结论:E-cadherin表达缺失在结直肠癌的发生、发展及转移过程中发挥重要作用;免疫组织化学和原位杂交2种方法在检测E-cadherin在结直肠癌中的表达上具有较高的一致性。 Objective: To discuss the expression and clinical pathological significance of E-cadherin in colorectal carcinoma.Methods: The expression of E-cadherin in 60 cases of colorectal carcinoma,20 cases of colorectal adenoma and 20 cases of paracancerous normal tissues were detected using immunohistochemistry and hybridization in situ.Result: The positive rate of E-cadherin protein in colorectal carcinoma(35%) was significantly lower than that in colorectal adenoma(70%) and paracancerous normal tissue(100%);The positive rate of E-cadherin mRNA in colorectal carcinoma(38.3%) was significantly lower than that in colorectal adenoma(75.0%)and paracancerous normal tissue(100.0%).The abnormal expression of E-cadherin were significantly correlated with the tumor differentiation,the TNM stage,depth of invasion,lymph node metastasis(P0.05),while not correlated with age,gender and the size of tumor(P0.05).Conclusion: The loss of E-cadherin plays an important role in the development and metastasis of colorectal carcinoma;Two techniques of immunohistochemistry and hybridization in situ have high consistency in detecting E-cadherin in colorectal carcinoma.
