The application of PCR-DGGE on the research of oral cavity microbial diversity

摘要 目的应用聚合酶链反应(PCR)-变性梯度凝胶电泳(DGGE)技术,分析健康人的口腔菌群及其菌种组成的多样性。方法抽取8名健康人唾液样本,分别提取唾液细菌总DNA,PCR扩增16SrDNA并进行DGGE和系统发育树分析。结果 8名健康人唾液样本的DGGE指纹图谱条带差异性较大,仅有65%相似性,表明采用PCR-DGGE对口腔微生物多样性的检测是可行的。结论 PCR-DGGE技术可较灵敏、直观地反映口腔微生物多样性情况。 Objective Polymerase chain reaction and denaturing gradient gel electrophoresis(PCR-DGGE) were applied to the microbial diversity examination of oral cavity.Methods Total DNAs were extracted from the plaques of 8 healthy people,16SrDNA of the bacteria was amplified and DGGE was also used to assess changes in bacterial composition from saliva microcosms from these 8 individual saliva samples.Results DGGE fingerprint patterns of saliva from 8 different donors displayed greater variability(65% concordance).PCR-DGGE method was a viable approach to examine the microbial diversity of oral cavity.Conclusion These results suggest that PCR-DGGE method is a sensitive and effective approach to examine the microbial diversity of oral cavity.
