摘要 目的:观察重组人神经生长因子(rhNGF)促进大鼠受损坐骨神经修复的作用。方法:夹闭大鼠坐骨神经造成挤压性损伤,给予rhNGF后不同时间点测定神经传导速度(NCV)和坐骨神经功能指数(SFI);同时采用大鼠坐骨神经切断再缝合法造成神经损伤,不同时间点测定动作电位潜伏期。结果:与模型组相比,4μg/kgrhNGF组在第35天、第56天的NCV显著加快(P<0.05),而8μg/kgrhNGF组在第35天、第56天的NCV非常显著加快(P<0.01);各剂量给药组与模型组的SFI相差显著,至第56天各剂量组SFI均恢复至手术前水平;rhNGF可使大鼠受损坐骨神经的动作电位潜伏期明显缩短,且显示量-效关系。结论:rhNGF对大鼠受损坐骨神经的愈合有促进作用。 Objective To observe the promotive effects of the recombinant human nerve growthfactors(rhNGFs) on repairing of injured sciatic nerve in rats. Methods: Rats' sciatic nerves were clippedand crush injuries were made. After being treated with rhNGFs, both the nerve conductive velocity(NCV) and the sciatic nerve functional index (SFI) were measured at different time. The cut-joint sciaticnerve injuries were made and the latency of action Potential was recorded at different time. Results: Incomparison with the model group, both the NCV of 4 μg/kg rhNGFs group and the NCV of 8 μg/kgrhNGFs group increased significantly(P < 0.05 and P < 0.01, respectively) on the 35th day and 56thday. The SFI of all rhNGFs groups decreased significantly as compared with the model group. Their SFIdid not reach the preoperative levels until the 56th day. The rhNGFs could evidently shorten the latencyof action potential of the injured sciatic nerves in rats. It could also show the dose-dependent manner.Conclusion: The rhNGFs have the promotive effects on the regeneration of the injured sciatic nerves inrats.
