IL-2基因工程菌生长密度对rhIL-2表达的影响

摘要 目的观察不同培养时间和细菌密度对重组人白细胞介素-2(rhIL-2)基因工程菌的收获量和rhIL-2表达量的影响。方法:用40L发酵罐进行了3批发酵试验。结果:工程菌于30℃培养4.5~5h,细菌生长密度(OD600)在1.7~2.0之间。在此条件下,工程菌再经42℃诱导培养4h,菌体的收获量和表达水平都比较理想,其湿重和表达量的均值分别可达6.3g/L(菌液)和31.6%。结论:诱导前不同培养时间和细菌密度能显著影响rhIL-2基因工程菌的产量和rhIL-2的表达率。 Objective To observe the influence of the different culture time and the density ofbacteria on the yield of the recombinant human interleukin-2 (rhIL-2) recombinant bacterial strain andthe rhIL-2 expression. Methods: Three lotS of fermentation tests were conducted in a 40 L fermentor.Results: After the interleukin-2 (IL-2) recombinant bacterial strain was cultured at 30℃ for 4.5 ~ 5hours, the growth density (OD600 ) of bacteria was 1 .7 ~ 2.0. As the recombinant bacterial strain wasfurther induced and cultured at 42℃ for 4 hours, the yield of the wet weight of bacteria and the rhIL-2expression would be more saisfactory. And the average wet weight of the bacteria and the rhIL-2 expres-sion were 6. 3 g/L (fermented liquid) and 31. 6 %, respectively. Conclusion: Before the induction, boththe different culture time and the bacterial density can significantly influence the yield of rhIL-2 recombi-nant bacterial strain and the rhIL-2 expression.
