子宫内膜异位症在位子宫内膜基质细胞体外蜕膜化模型的改良

摘要 目的:通过对目前常用的子宫内膜基质细胞体外蜕膜化模型进行改良,建立更准确和高效的子宫内膜异位症体外细胞模型,了解子宫内膜异位症患者在位内膜的蜕膜化能力,进而研究子宫内膜异位症与不孕的关系。方法:将子宫内膜异位症患者在位内膜细胞随机分为三组,第一组常规用E2(10nM)和P4(1μM)作用96 h(EP1组),第二组常规用E2(10nM)、P4(1μM)作用14 d(EP14组),第三组改良组用E2(10nM)、P4(1μM)和8-溴-cAMP(0.5mM)作用96 h(EPC组),收集细胞后测定PRL(催乳素)和IGFBP1(胰岛素样生长因子结合蛋白1)的mRNA水平。结果:EPC组作用96 h后细胞形态发生明显变化,PRL和IGFBP1明显高于EP组。结论:与常规模型相比,8-溴-cAMP联合P4、E2能明显促进子宫内膜异位症在位子宫内膜基质细胞体外蜕膜化进程,使在位内膜体外蜕膜化能力明显优于常规体外模型组。
